Book Description: Excerpt from Interaction of Aziridines With Cellular Components During the past 30 years much work has been done on'the mechanism of action of biologically active alkylating agents, and the results of this work have made evident the extreme complexity of the topic. Nitrogen mustards dominate much of the work on biological effects of alkylating agents, with other compounds belonging to the same class aziridines, sulfonic acid esters, and epoxides-appearing less frequently. Nevertheless, broad generalizations about the biological effects and biochemical mechanisms of all biological alkylating compounds have been and still are frequent. During World war I and the postwar period, workers (lynch et al. 1918, Pappenheimer and Vance 1920, Flury and Wieland 1921) investigating the vesicant properties of mustard gas realized that it and related chemicals possessed unusual biological properties. Marshall (1919) proposed that the action of mustard gas was due to its rapid penetration into the cell, hydrolysis within the cell to generate hydrochloric acid, and subsequent destruction of some mechanism or part of the cell by hydrochloric acid. However, Peters and walker (1923) demonstrated that there was no correlation between the rates of liberated acid and the vesicant action of mustards. In the midthirties.
