Book Description: Recently, autophagy-related genes polymorphisms have been implicated in several autoimmune diseases including systemic lupus erythematosus, rheumatoid arthritis, psoriasis, and multiple sclerosis. Autophagy is the highly conserved protein degradation pathway from yeasts to humans and is essential for removing protein aggregates, misfold proteins in healthy cells. More and more studies reveal autophagy and autophagy-related proteins also participate in immune regulation, such as removal of intracellular bacteria, inflammatory cytokine secretion, antigen presentation, and lymphocyte development. The generation of autophagy-related protein knockout mice advances our understanding of autophagy in the immune system and autoimmunity tremendously. For instance, autophagy-related proteins deficient mice are protected from experimental autoimmune encephalomyelitis, an animal model of multiple sclerosis, and autophagy-related proteins deficient mice are protected from TNF[alpha] joint destruction in experimental arthritis. One of the links between autophagy and autoimmune diseases is activation of nuclear factor [kappa]B (NF-[kappa]B). Autophagy has been shown to regulate NF-[kappa]B activation while NF-[kappa]B activation often leads to autoimmune disease pathogenesis. In this article, we explore the role of a novel autophagy-related protein WDFY3 in NF-[kappa]B activation and an animal model of autoimmune diseases. More detailed understanding the role of autophagy and autophagy-related proteins is needed for developing therapeutic interventions target autophagy pathway/autophagy proteins for autoimmune diseases.
