Book Description: The availability of the genome sequence and a genetic map of the northern root-knot nematode, Meloidogyne hapla, make this nematode an excellent system for investigating traits important for plant parasitism. The mode of reproduction in most isolates of M. hapla is facultative meiotic parthenogenesis, a novel reproductive mechanism which makes this species amenable for selfing and outcrossing. Comparing meiotic progression during germline development in the gonads of mated and unmated M. hapla strain VW9 females has identified differences in meiotic mechanism and in early embryonic development in the same species reproducing via sexual and asexual modes. A preliminary genetic map was produced by analysis of segregating AFLP markers in 183 F2 lines derived from parental strains VW8 and VW9. I have used SNP markers to map new loci in the F2 population with the Illumina Golden Gate assay system. These polymorphisms were identified by obtaining genomic sequence from pooled F2 progeny using an FLX 454 sequencer. This has allowed positioning of sequence contigs on the genetic map and to determine genomic organization of candidate genes for parasitism. A forward genetics approach was used to dissect parasitism related traits by utilizing F2 lines from the same cross (VW8 x VW9) used in the genetic map construction. Differences in attraction to wild potato Solanum bulbocastanum, in ability to reproduce and in galling severity on this host were identified as quantitative trait loci (QTLs) and were mapped to linkage groups.
